Ready to use Silica Clean-up Columns Following Organic Synthesis

Introduction
Following organic synthesis, reaction mixtures contain a range of by-products, excess reagents and starting materials as well as the target molecule(s). Chemists routinely use a range of techniques for cleaning up these complex mixtures – one of the simplest options just became even more straightforward. The use of lose or bulk silica and a Buchner Funnel allows the removal of much of the polar impurities and “tar” formed as part of the reaction.
Ready to use Silica Column & Processing Station

TELOS Silica open tubular or SPE format disposable columns provide an easy solution to this very common process – no need for washing and drying of Buchner Funnels – place the column on a processing station, filter the sample and then move onto to the next stage of the purification or work-up. The filtrate is then taken to the next purification technique. No need to prepare and wash Buchner Funnels anymore.
Solvent Systems
If certain desired compounds are retained by the TELOS Silica Column, wash with a solvent compatible with the subsequent flash chromatography separation. The following table provides recommendations for suitable TELOS Flash Silica Column wash / elution solvents:
| Flash Chromatography Solvent System | TELOS Flash Silica Wash / Elution Solvent* |
| n-Hexane, iso-Hexane or Cyclohexane/Ethyl Acetate | Increase polar solvent by 15% from optimum TLC solvent system (Rf of target compound is ~0.5) |
| DCM / Methanol | Increase polar solvent by 5% from optimum TLC solvent system (Rf of target compound is ~0.5) |
*use 2-3 column volumes to elute; a 10g Silica has a column volume of ~14ml
Capacity
TELOS Flash Silica Columns have a capacity (w/w) to flash chromatography columns. The TELOS Flash Silica Column can be used as a “pre-column” preventing much of the “tar” or polar impurities and by-products from reaching the top of the flash column. This increases the capacity and separating efficiency of the main flash column.
