Introducing TSKgel UP-SW3000 SEC columns

Antibody therapeutics are enjoying high growth rates, with the major areas of therapeutic application being cancer and immune/inflammation-related disorders including arthritis and multiple sclerosis. In 2013, six of the top ten best-selling global drug brands were monoclonal antibodies (mAbs) and more than 400 monoclonals were in clinical trials. The characterization of these complex biomolecules is a major challenge in process monitoring and quality control. The main product characteristics to be monitored are aggregate and fragment content, glycosylation pattern and charged isoforms. Our new TSKgel UP-SW3000 SEC columns from Tosoh Biosciences are the latest in a long line of tools to to allow characterization of mAb isoforms.

Unparalleled Resolution

Offering unparalleled resolution in the trimer/dimer/monomer/fragment region, these new 2μm columns provide rapid, high resolution separation of mAb samples. TSKgel UP-SW3000 columns are specifically designed for the analysis of mAbs and other biopharma products and can be used on both HPLC and UHPLC systems. The columns are packed with 2 μm silica-based beads shielded with a hydrophilic diol-type bonded phase that prevents the silica surface from interacting with protein samples to ensure excellent peak shapes.

Cost-effective and fast

As well as offering state-of-the-art separating power these columns also represent a cost-effective alternative to existing SEC methods – one TSKgel UP-SW3000 column can provide superior separation, in less time than two TSKgel G3000SWxl columns in series. TSKgel UP-SW3000 columns feature the same pore size as the well-established TSKgel G3000SWxl columns, hence methods developed using TSKgel G3000SWxl columns can easily be transferred to TSKgel UP-SW3000 columns on UHPLC systems as well as on conventional HPLC systems.

As with all TSKgel SEC columns, TSKgel UP-SW3000 columns offer superior reproducibility injection-to-injection, from column-to-column, within the same lot and from lot-to-lot. %RSD values for peak parameters including retention times, area peak height, peak asymmetry, and theoretical plate count demonstrate the exceptional reproducibility of these columns, making them a proven platform for mAb analysis.
